Current Research
My research focus areas are on various aspects of herbal medicines. I have worked on bioactivity guided investigations, mutagenic activity investigations, as well as antimicrobial, wound healing, and anti-helminthic activity investigations. Standardization, monograph development and ethnobotany of herbal medicines are still ongoing.
1. Wound healing activities of Ghanaian medicinal plants on skin cells
This study investigated the antimicrobial and wound healing activities of Clerodendron splendens G. Don (Verbanaceae) in both in vitro and in vivo experimental models. A number of Ghanaian medicinal plants were also investigated for their wound healing activity on keratinocytes and fibroblasts (skin cells). The most promising plant (Combretum mucronatum), was selected for in-depth phytochemcal characterization and in vitro wound healing activity studies. The wound healing potential of crude extracts, fractions and isolated compounds were studied. A novel ICH-validated Ultra High Pressure Liquid Chromatographic-Mass spectrometric (UHPLC-MS) method for the analysis of the major constituents (epicatechin, procyanidin B2, vitexin and isovitexin) of this extract was also developed. This project was funded by the German Academic Exchange Service (DAAD) from 2011 to 2014.
2. Herbal medicines used in the treatment of typhoid
This project involved the appraisal of these commercial products, determination of their microbiological quality, and investigations into their anti-Salmonella activity against various Salmonella enterica serovars.
3. Ethnopharmacology/Ethnobotany
This project involved a survey of commonly sold medicinal plant materials at major markets in Accra and investigations into their mutagenic activities. Results identified traditional herbal plant materials that are commonly sold amongst Ghanaians living within the Accra metropolis; and further conduct preliminary in vitro mutagenic investigations on the herbs. Findings resulted in identifying and documenting plant materials; the way and manner by which these plant materials are obtained, prepared and administered. Samples of the named plant materials were collected for the purposes of identification, documentation and in vitro testing for mutagenic activity. The resultant crude extracts have been assessed for their in vitro mutagenic activity. This mutagenic toxicological study identified herbal materials that have the propensity of causing mutations, and hence the likeliness of being linked to mutagenic diseases like cancers and resultant organ failures. This study was funded by an Investigator Led Grant from the University of Ghana. Experience from this study was also translated within a collaborative framework to an ethnobotanical study in Benin.
4. Quality control of commonly used crude drugs in Ghana
This study investigated a number of widely patronized herbs on the Ghanaian markets (Hibiscus sabdariffa, Moringa oleifera, Kyaya senegalensis, Alstonia boonei, Phyllanthus amarus etc.) for various characteristics that can be used in determining their efficacy, identity and quality to safeguard the Ghanaian consumer.
5. New insights into the molecular mechanisms of anthelmintic proanthocyanidins
Eight out of the 18 neglected tropical diseases (NTD) covered by the WHO are caused by helminths. The main strategy to control and eliminate NTDs is preventive chemotherapy. Many of the currently available drugs have been around for quite a long time, and the drastic increase in the development of anthelmintic resistance has also become a major problem in veterinary medicine. The aim of the study was to define the underlying mode of action and the molecular target of proanthocyandins, which is in principle is unknown until now. Based on these mechanistical studies the respective effects will be correlated to the outcome of in vivo infection studies with different intestinal parasites in dogs and sheep as well as to human parasites, investigated using ex vivo approach. This study is still ongoing and is being conducted within an African-German network. I am the lead investigator working on the phytochemical and analytical studies of work package III of the above study. This project is funded by the Deutsche Forschungsgemeinschaft (DFG).
